LABORATORY:
after the evaluation, the ejaculate is blended with the chosen
extender that must be previously prepared at a temperature ± 1ºC
regarding to ejaculate's. For this purpose,
both extender and ejaculate must be placed into a water bath
at 35ºC. Once the
ejaculate has been diluted, it is left at room temperature
(20-22ºC) for at least 2- 3 hours in order to achieve 20ºC
gradually. Then it is placed into the fridge at 15ºC.

PRESERVATION:
in the 15ºC fridge the semen doses will remain a period of time
that may vary depending on insemination demands of, ejaculate's
quality and extender used. During all this
period, temperature must not decrease below 15ºC. controlling it
through a maximum-minimum thermometer into the
device.

INSEMINATION:
before insemination, semen doses are removed from the 15ºC
fridge and are placed into a water bath at room temperature
(20-22ºC). After 10-15 minutes, the water bath is switched on
to get a final temperature of 35ºC along 10-15 minutes, so doses go from 15 to 35ºC
at least during 20-30 minutes. Once at 35ºC, this temperature
must be maintained without changes until the time of
insemination. However, a long lasting period should be avoided
since spermatozoa would become exhausted and
die.

very important to remember that temperature decrease of to 15ºC
must be gradual, because we submit the spermatozoa to a temperature
against its physiology that provokes that plasma membrane
becomes rigid and Dose heating can be faster the spermatozoa go again to its physiological temperature.
Temperature control is also important during the evaluation of semen motility. Boar spermatozoa get all its motility potential
at 37ºC, therefore being lower at 35ºC the motility will be lower. In order to
assure a correct evaluation of sperm motility, we must place a
thermometer "right clean" directly into the collection
vessel. On the other hand, the microscope plate is at room
temperature so that reason the evaluation must be as quick as
possible, just at first sight. If we have a warm plate, we must
check that surface temperature is 37ºC. Termometer with digital thermometer.
